I know people have mentioned it before, but I feel the need to reiterate the power of spaced repetition systems (SRS) for those who have yet to discover them. It helps you learn more quickly and efficiently! Quite simply, it's like a flash card application, only smarter in that you grade yourself on how well you answered a question and the program will schedule the next best time in which the card will appear for review again. Basically, things you don't now well appear more often than things you know well so you don't waste time.
The two best free SRS programs out there are Mnemosyne (http://www.mnemosyne-proj.org) and Anki (ichi2.net/anki/).
The most effective method I've come across in my Japanese studies is to learn LOTS and LOTS of SENTENCES in conjunction with an SRS.
Khatzumoto's blog, All Japanese All The Time (http://www.alljapaneseallthetime.com) describes the SRS/sentences method in greater detail and he also provides a lot of motivational posts.
On a side note, if you own an iPhone or iPod touch, you can use iSRS (http://groups.google.com/group/isrs-support) for Mnemosyne or iAnki for Anki (http://ichi2.net/anki/wiki/iAnki) to do your learning on-the-go!
